Synthesis and In vitro Cytotoxicity of a Novel Efficient Cisplatin-loaded poly N-butyl cyanoacrylate

Abstract:

Purpose: Typically, drug-loaded polymer colloidal carriers are synthesized by the drug entrapment during anionic emulsion polymerization. The purpose of this study was to trial the ability of Cisplatin-loaded Poly Butyl Cyanoacrylate (PBCA) nanoparticles. Methods: The cytotoxicity of Cisplatin-loaded PBCA nanoparticles was evaluated by MT assay. The Polymeric nanoparticles have been characterized using TEM, SEM, FTIR and DLS. Results: Polymeric nanoparticles with loaded and unloaded drug prepared by this method, have spherical structure with 350 and 230 nm in diameter and their zeta potentials were -3.2 and -10.7 mv respectively. The average amount of drug loaded on the nanoparticles was 70.88 ± 5.51%. MTT assay was carried out to evaluate the cytotoxicity property of the nanoparticles. Inhibition concentration values (IC50) for Cisplatin-loaded Nanoparticles were 20.8 and 18.2 μM for 24 and 48 h respectively. Conclusion: It was shown that cisplatin-loaded nanoparticles strongly increased cytotoxicity in comparison to classic drug in the MCF-7 cell line.
