Nimodipine is antihypertensive drug with an oral bioavailability of only 40% with short half life as 1-2 hrs.The goal of present investigation, an attempt has been made to formulate sustained release matrix tablets of nimodipine by using various natural matrix former gums as Xanthan gum, Olibanum gum, and Locust bean gum separately. Tablets were prepared by wet granulation method. for uniformity of weight, hardness, friability, thickness, content uniformity, In-vitro dissolution, drug-excipients interactions, swelling index study also carried out. Among different formulation,F-9 which contain Locust bean gum with drug: polymer ratio as (1:3) and (46.15 % gum) exhibited precise controlled release of drug up to (88%) within 10 hrs time period. The effect of other parameter such as speed of rotation, types of dissolution apparatus was also studied for F-9 but statistically there was no significant difference observed in dissolution when data is analysed by one way ANOVA (P<0.05 ) and Student t- test. In addition Similarity factor f ) above 50 support similar release profile. 2 0 Optimized formulation F-9 was subjected to stability studies for three months at 45 c with 75±5% RH as per ICH guidelines and result does not shows any changes in physical parameters and in-vitro release studies. Majority of designed formulations displayed nearly zero order release 2 kinetics (R =0.9925) and Korsmeyer and Peppas equation give release pattern with value of (n=0.6054) indicating non-fickian or Anomalous types of diffusion take place through matrix of Locust bean gum. The results demonstrate the feasibility of natural gum in the development of matrix tablets for controlled delivery of nimodipine.
